Any algorithm used to reconstruct the 3D location of each individual molecule must take into account the emission signal from all fluorophores detected, including those that are not part of a molecule. Ideally, a molecular reconstruction algorithm should be able to separate PSFs that are not part of a molecule, because these are not representative of the real 3D structure of a molecule, from those that are. Existing algorithms often incorporate a metric that assigns a score to the likelihood of each detected PSF being real. The main difficulty with this approach is that the ‘real’ structure of each molecule is not known, so the scoring metrics usually rely on comparison of each detected PSF to a model PSF, such as a Gaussian. This model is either based on an average molecule in a 3D reconstruction or a theoretical model that calculates the intensity pattern expected from an ideal molecule. Optimisation of the model is therefore an important element of molecular reconstruction algorithms and is related to the number of fluorophores and the SNR. In addition, algorithms often rely on local maxima within the background-corrected image to obtain the candidate molecule locations. These locations can differ from the peaks identified during detection, and the choice of the ‘correct’ location is a function of the SNR and how large the region of the image that is considered. The higher the SNR, the more likely there will be a local maximum that corresponds to the true molecule position, in contrast to a chance fluctuation of the background.

I am running a PC with Windows 7 and Adobe Photoshop CS6. I am wondering how to end the trial period and convert to a full copy of Photoshop CS6. I tried closing down Photoshop, which produced a message telling me to return the disk. I did that, but the same message returned. How can I end this?

Hi Dave,I was able to get my English Indesign CS6 to switch to German. But there is one Problem. The moment I startet Indesign there is the error message that my trial version ends soon altough I bougt a regular Version with correct serial number by an adobe seller. Can anybody help me? 827ec27edc
